tylikSo I ran an agarose gel with my PCR products from yesterday. And... it kind of worked.
The parts related to prep all worked. My bands were very clear and defined. But I didn't get any products at all from the mutants, which means something was wrong with one of the primers, which probably means that somewhere along the way what we have isn't quite what we think we have. Hopefully not in too terrible of a way.
Um. Data curation. Sticking pins in eyes. Yes.
Meanwhile, in terms of checking drug resistance and such it looks like I have a whole bunch of happy double mutants. (I wouldn't exactly call them healthy.) I am going to be really unhappy if it turns out the are not the right double mutants. (This is fairly unlikely. The deletions are probably right, they might have been done differently than we thought.)
(I really need to get around to making a yeast icon.)
